A gene encoding one of the proteins involved in the DNA replication has been inactivated by a mutatiion in a cell. In the absence of this protein the cell attempts to replicate its DNA for the very last time. What DNA products would be generated in each case if the following protein were missing / mutated? (consider each one individually, not in combination and briefly explain your reasoning. in other words, what would happen if this enzyme didn't work? What would be produced in replication
(a) primase
(b) DNA ligase
(c) DNA polymerase I
a) Primase- during DNA replication, primase make short RNA, where the nucleotides are complenetary with the single strand DNA. Now, to start DNA replication, DNA polymerase requires the presence of an intial RNA / DNA primer. Thus without primase, there will be no DNA replication.
b) DNA ligase- DNA ligase is required to join the OKAZAKI fragments, at the time of DNA replication.. Thus without DNA ligase, Okazaki fragments of the DNA lagging strand will not joined and as a result new DNA strand will not be synthesized.
DNA polymerase I- during replicaiton, DNA polymerase I move out the RNA primer and replace with DNA nucleotide. This is known as exonuclease activity of DNA polymerase I. Thus without DNA poymerase I the lagging strand will not synthesized which will also inhiit DNA replication.
