Restriction digestion is the process of cutting DNA molecules into smaller pieces with special enzymes called restriction endonucleases. If they cut DNA molecule into small fragments then the small fragments will run faster into the gel. They cover large distance, and we will see the bands in the upper portion of the gel. If they cut DNA molecule into large fragments then the large fragments will run slower into the gel. They cover small distance, and we will see the bands in the lower portion of the gel.
