Sanger sequencing is a method of in vitro DNA sequencing. It is based on the selective function of dideoxynucleotides in chain-terminating during DNA replication.
The DNA sample is divided into four separate sequencing parts, Each part has one dideoxynucleotide (ddATP, ddGTP, ddCTP or ddTTP). Dideoxynucleotide has different density than normal nucleotide. Different dideoxynucleotides give different flouriscence which indicate chain termination of a particular type. When exposed to the x-rays, the dark bands correspond to DNA fragments of different lengths. A dark band in a lane indicates a DNA fragment that is the result of chain termination due to presence of dideoxynucleotide . The relative positions of the different bands among the four lanes from bottom to top is used to read the DNA sequence.
