How can TLC be used to determine the best solvent or combination of solvents for a certain application?

Homework Answers

Answer #1

TLC can be mostly used in the synthetic labs for monitoring the reactions. We have to use proper solvent or combination of solvents for the seperation.Normally TLC playes made by a layer of silica or alumina which serves as a stationary phase and the mobilephase we choose by oir choice or depending on the polority of the compounds we spotted on the TLC plate. Dependong on polority diiferent compounds will moove different distances on the plate. For example if we spotted polar compound we have to use

1. Methanol and DCM combination

2. Methanol and Chloroform

For non polar compounds

1. Ethylacetate in hexane

2.acetone - hexane

3. DCM- ethylacetate

4. Toluene

1. Several different combinations of solvents have been used for TLC so far. For example, 80% ethyl acetate/20% hexanes was chosen for the TLC separation of analgesics. Consider that solvent for the following questions. a. Is it important that the solvents are miscible? Briefly explain why or why not. b. Briefly explain what would be different if the ratio was flipped (i.e., 80% hexanes). c. Would 100% hexanes have made a good solvent? Briefly explain why or why not. d....

Ethyl acetate is another common organic solvent used in the type of TLC experiment you performed: it has a higher molecular weight and boiling point than either petroleum ether or diethyl ether, and is more polar than them as well. If ethyl acetate had been used as the mobile phase for a TLC analysis of F and AF, how would their Rf values compare to those found using the 1:1 solvent mixture you used? That is, predict how the behavior...
