Column chromotography lab question?

Once the chromatographic column has been prepared, why is it important to allow the level of the liquid in the column to drop to the level of the alumina before applying the solution of the compound to be separated?

Answer #1

In wet packing of chromatographic coloumn we use eluent to wet the entire coloumn before the application of compound. It is very important to drain the excess eluent which is in above the stationary phase. Why it is important is our compound which has much more non polar compound can be dissolved in the eluent and hence it can affect separation of individual compounds. Also if you add compound with excess of solvent the compound will not adsorbed over Silicagel/alumina hence it will leads to improper separation of bands.

Question: On the Level Construction Company has been growing and needs to purchase a new machine. They have been researching multiple companies and have narrowed down their choices to two options - based on reliability and customer service. They now need the help of accounting to determine which option is better financially. Company 1 - Millipede The equipment would cost $3,000,000 up front. It has a 12 year useful life. Millipede's equipment would allow for 2 additional construction jobs per...

Lab 9 – Molecular Biology In this lab, you will prepare an agarose gel and use gel electrophoresis to compare the size of 2 dye molecules Methyl orange and Ponceau G. You will also analyze an “unknown” sample that contains a mixture of two dyes. Dye molecules with lower molecular weight or greater electrical charge will migrate faster through the gel, than dye molecules with greater molecular weights or lesser electrical charge. Materials: Mini gel electrophoresis chamber 6 Tooth comb...

A solution of a theoretical triprotic acid was prepared by dissolving 4.037 g of solid in enough DI water to make 500.0 mL of solution.   10.11 mL of a 0.592 M solution was required to titrate 20.00 mL of this acid's solution. Part A What is the concentration of the acid solution? Part B What is the molar mass of the acid? Hint: You need to calculate the total moles in the 500.0 mL solution (the full 500.0 mL was...

Below is the background info for the lab assignment. The 4 blank boxes are the questions I would like answers too. The end point for the fine titration was 35.90 mL in the burette when the solution turned bright green again. And the coarse titration I got 35.36mL as the end point. 35.9 mL is exact in case you need that info. Thanks! Background In this lab, we will determine the amount of alcohol (ethanol), C2H5OH, in a commercial vodka...

I have provided the information for the lab below. I want to see the chemical reactions that take place during each step of the experiment. So in the procedures when a chemical was added i would like to see what the products and byproducts that were formed. I also want to see one over all reaction for this experiment to just get an overview of what exactly is happening during the experiment. Thank you:) Part B: Synthesis of [Co(NH3)5Cl]Cl2 Objective:...
