In column chromatography, one the chromatographic column has been packed, why is it important to drain the level of the liquid in the column to just above the top of the column of silica gel before applying the compound to be separated?
This is required to avoid the solubility of compound (which is to be seperated) through out whole solvent which is present excess in the column. If solvent is present in excess, then there are chances that mixture of compounds will be distrributed in the excess solvent present in column due to some solubility and due to this issue, compund will start comming covering a broader region in silicagel instead of a narrow band which is required for better separation.
