What are some limitations to a serial dilution lab? As well as possible errors and how they can be avoided?

Answer #1

Answer :

Many laboratory protocols requires serial dilution of reagents or compounds.

Serial dilution faces major challenges.

Error propagation across columns or rows. They are procedural errors, by inaccurate transfer, dilution and inadequate mixing.

Result is highest dilutions will have maximum error.

This error can be compensated by longer mixing times are required. This increases time required to do serial dilution procedure.

With raising of mix tip height precision increased.

Mix liquid class setting modification with mixing velocity increased accuracy.

Dynamic tip extension and retraction also improved precision. Tips dipped more depth and retracted during dispensing.

Serial dilutions Please draw how to prepare this serial dilution prepare a series of p nitrophenyl phosphate concentrations from 1x10^2 M from to 3.125x10^-4 using the DEA buffer as diluent. do this by srial diluting the 1x10^-2M stock by 1/2 so you end up with 2 ml each of p-nitrophenyl phosphate concentrations of 5x10^-3M, 2.5X10^-3M, 1.25x10^-3M, 6.25x10^-4M and 3.125x10^-4M

Laboratory experiments, regardless of how well-equipped and well-managed they are, are always subject to limitations and their findings don't 100% match up with theoretical frameworks. Give a few examples as to what limitations and considerations we need to keep in mind to validate an equation or theory.
