After a restriction digestion of a TOL plasmid, why should the enzyme be inactivated immediately after the restriction experiment? How can the inactivation be achieved?

Homework Answers

Answer #1

Immediately after the restriction experiment the restriction enzymes should be inactivated immediately by treating with inactivation temperature (heat treatment) at which the particular used restriction enzyme inactivated . These RE should be immediately inactivated because RE enzymes have a activity time for proper restriction but if their activity remain activefor long time than they can cause overdigestion of plasmid and multiple fragments of TOL plasmid will obtain that is not the result we want.

You digest a plasmid with a restriction enzyme that recognizes a single site on the plasmid. When you perform gel electrophoresis on the digestion product, you quickly realize that there are two bands; one at the expected size and one near the well. Which of the following best explains the outcome? DNA was trapped in the agarose gel The presence of the chromosomal DNA Some plasmids were not digested The some plasmids ligated together

Restriction enzymes, also known as restriction endonucleases, are enzymes that cut a DNA molecule at a particular place. They are essential tools for recombinant DNA technology. The enzyme "scans" a DNA molecule, looking for a particular sequence, usually of four to six nucleotides. Once it finds this recognition sequence, it stops and cuts the strands. This is known as enzyme digestion. On double stranded DNA the recognition sequence is on both strands, but runs in opposite directions. This allows the...

Why are type II endonucleases, instead of types I and III, preferably used in molecular biology? A. Endonucleases I and III use ATP and cut away from their recognition sites, but an endonuclease II enzyme does not use ATP and cuts within the recognition sequence B. Endonucleases I and III cut at only a few DNA sequences, whereas endonuclease II enzymes cut at a very large number of recognition sequences C. Endonucleases I and III are large, which causes highly...

Precursor oligosaccharides are transferred to the asparagine (N) immediately after that amino acid has reached the ER lumen during protein translocation. They are used as tags to mark the state of protein folding. By checking these tags, the ER determines which proteins are folded properly and can leave the ER. When and how is the protein targeted for degradation? Briefly explain in your own words how the ER “knows” that protein should be degraded.

Answer the following questions: Why should the glassware be dry for this experiment? Why might water contamination be a problem when using the reagent acetic anhydride; what byproduct might form due to moisture? 1a This reaction can easily be “followed” by TLC, i.e., as the reaction proceeds, salicylic acid starting material is consumed, simultaneously with formation of product. Curiously the SM will have an Rf value higher than the product. Why is this odd? ? (What structural feature in the...
