What would you expect to happen to the retention time of the Capillary Zone Electrophoresis (CZE) experiment if you decreased the run voltage? Capillary length? Increase the buffer viscosity?

Homework Answers

Answer #1

If the run voltage increases the retention time decreases, as the velocity of ions is directly proportional to the velocity gradient, so as the run voltage increases the separation takes place faster and hence the retention time decreases.

If the Capillary Length decreases the retention time also decreases and vice-versa, Because longer the tube, longer is the time taken for separation and hence longer is the retention time and vice-versa.

If the viscosity of Buffer decreases ,the retention time also decreases because the resistance offered against the movement of ions gets reduced and so the separation happens faster and hence the retention time reduces and vice- versa
