Which compound and why would you expect to have a shorter retention time on a reverse-phase HPLC column with a water/acetonitrile mixture adjusted to pH3 with acetic acid — benzylalcohol or benzoic acid.

Homework Answers

Answer #1

In reverse phase HPLC column, the stationary phase is nonpolar. Between, benzyl alcohol and benzoic acid, benzyl alcohol is less polar as compared to benzoic acid. Less polar benzyl alcohol is thus going to have more attractive intermolecular interaction with the stationary phase as compared to benzoic acid. Hence, benzoic acid will elute first when eluted with water/acetonitrile mixture adjusted to pH 3 with acetic acid . Therefore, benzoic acid is expected have a shorter retention time.

In a normal-phase partition column, a solute was found to have a retention time of 29.1 min, and an unretained sample had a retention time of 1.05 min when the mobile phase was 50% by volume chloroform and 50% n-hexane. calculate a) k for the solute and b) a solvent composition that would bring k down to a value of about 10.
