Different Media types:-

1) Broth= a liquid medium. Advantage: tube is easy to store and transport.

Disadvantage: can’t see colony morphology.

2) Slant= tube of solid medium at an angle. Advantage: tube is easy to store and transport, can see colony morphology.

Disadvantage: small surface area.

3) Agar deep= tube of solid or semi-solid medium. Good for organisms that prefer reduced O2 and to evaluate motility. Bacteria that have a flagellum , and are motile will migrate from the inoculation point through the medium.

Disadvantage:- Because oxygen will with time diffuse into the Agar Deep tubes after they have been made, if they have set for a long time, it is recommended that they be remelted to remove the oxygen.
