Answer #1

Advantages:

1) Liquid culture
  • This method can be standardized with more efficiency.
  • Can tell whether the bacteria is aerobic or anaerobic, as aerobic ones need oxygen and colonise at surface . But anaerobic ones colonise at lower end. (Sensitivity).
2) pour plate
  • It can determine the bacterial concentration , without the need of any previous culture.
  • Can give precise results .
3) streak plate:
  • Helpful in isolation of separate pure colony from a mixed bacterial culture.
  • Easy to perform, is less complex.
4) slants
  • Provides larger surface area for growth of bacteria.
  • Easy to store and maintain.
  • Can be stored for longer periods.

You and your lab partner wish to determine the concentration of a bacterial broth culture. You dilute the culture by 1:1,000,000 (or 10-6). You then pipet 1 ml into a Petri dish and pour melted TSA on top of it. After gently swirling to mix and allowing it to harden, you incubate overnight at 37° C. The next day you count 95 colonies on the plate. What is the concentration of the original culture in CFU/ml? How would your answer...

A grad student has a culture of E. coli with a total volume of 600 mL, and OD reading at 550 nm of 1.7. He makes serial dilutions of the culture, starting with a transfer of 1 mL culture into 9 mL tryptone broth. Next, he transfers 0.1 mL of this dilution into 0.9 mL tryptone broth, and repeats this two more times to make a total of 4 serial dilutions. He then pipets 200 uL from the fourth dilution...

A tissue culture plate experiment is done by adding 2 drops of hydroxylamine solution, 7 drops of ligand solution, and 3 drops of iron solution to a well in the plate. If you want to repeat the experiment on a larger scale, what is the conversion factor from plate to cuvette assuming that you need to make 2.8 mL of total solution volume?

Could you change to paragraph Yeast culture(Plate Set-up) Label plate with a sharpie.(A= Amber Lens, L=Blue Lens) Pipette 0.15mL of yeast solution on to the agar. Tilt and rotate the plate. Use disposable L-rod spreader to spread the liquid so that the agar surface is evenly covered. Place the plate in a dark place for 10-20minutes until the agar no longer appears wet. Cover “foil” half of the plate with aluminum foil. Expose the plate to a UV source for...
