Yeast culture(Plate Set-up)

1. Label plate with a sharpie.(A= Amber Lens, L=Blue Lens)
2. Pipette 0.15mL of yeast solution on to the agar.
3. Tilt and rotate the plate. Use disposable L-rod spreader to spread the liquid so that the agar surface is evenly covered.
4. Place the plate in a dark place for 10-20minutes until the agar no longer appears wet.
5. Cover “foil” half of the plate with aluminum foil.
6. Expose the plate to a UV source for the assigned length of time. Be sure to position the plate so that the exposure is direct.
7. Remove the foil from the plate.
8. Place the plate upside down in a 30 degree Celsius incubator (1~2 days total). After that, plates will be put in a refrigerator.

Yeast Culture plate Set-up:

Prepare agar plates for yeast culture and label them with a sharpie. Pipette out 0.15 ml of yeast solution on to the agar plate. Now, tilt and rotate the plate and spread the liquid on the agar surface, evenly, using a disposable L-rod.

Place the plate in the dark for 10-20 minutes until agar is no longer wet. Cover half of the plate with an aluminium foil and expose the plate to a UV source for certain time. Be assured that the position of the plate is directly exposed to UV. Remove the foil from the plate and place it upside down in an incubator at 30 degrees celsius for about two days.

After two days, place the plates in a refrigerator for further storage.
