Yeast Culture plate Set-up:
Prepare agar plates for yeast culture and label them with a sharpie. Pipette out 0.15 ml of yeast solution on to the agar plate. Now, tilt and rotate the plate and spread the liquid on the agar surface, evenly, using a disposable L-rod.
Place the plate in the dark for 10-20 minutes until agar is no longer wet. Cover half of the plate with an aluminium foil and expose the plate to a UV source for certain time. Be assured that the position of the plate is directly exposed to UV. Remove the foil from the plate and place it upside down in an incubator at 30 degrees celsius for about two days.
After two days, place the plates in a refrigerator for further storage.
