A short tandem repeat (STR) is used to compare specific loci on DNA from two or more samples. It is a microsatellite, consisting of a unit of two to thirteen nucleotides repeated hundreds of times in a row on the DNA strand. This method measures the exact number of repeating units. In this method, probes are attached to desired regions on the DNA, and a polymerase chain reaction (PCR) is employed to find the lengths of the STR.

STR is used in forensic analysis to evaluate the presence of specific STR loci on nuclear DNA. Forensic science enables scientists to distinguish two or more DNA samples. The DNA profiling is based on short tandem repeats. Unrelated people have almost different numbers of repeating units and STR can be used to discriminate between unrelated individuals.
