How can I explain QF-PCR technique procedure steps for prenatanal diagnosis?

Homework Answers

Answer #1

QF-PCR

QF-PCR is Quantitative Fluorescent Polymerase Chain Reaction, which uses fluorescent labelled primers to amplify STR regions from DNA, by PCR. STR (short tandem repeats) are highly polymorphic sequences found in human DNA. The amount of Fluorescent labeled amplicons were measured after fragment length separation in capillary electrophoresis.

QF-PCR is a quantitative method that determines the presence of different alleles. With this method, the chromosome copy number can be determined. Thus, this diagnosis of fetal chromosome ploidy can be performed within 4-5 hours.

Steps :-

  • DNA extraction from amniotic fluid.
  • PCR amplification.
  • Capillary Electrophoresis.
  • Export of data
  • Analysis or diagnosis of PCR product.
