Describe the steps and their purpose in the thermal cycle of PCR.

Homework Answers

Answer #1

Steps of thermal cycle of PCR :

Thermal cycling is a process where polymerase chain reaction involves in heating and cooling by thermocycler machine .It has 20-40 series of repeated cycle. In this cycle ,each cycle there are 3 steps found. The steps are given below .....

  • Denaturation - here, double stranded template DNA heated into 94°C and then separate into two single strands .
  • Annealing - here, lower temperature that is 50° C enable DNA primer to attaching thr template DNA.
  • Elongation - here, new strand DNA is made by raising the temperature of 72°C. Purpose of thermal cycle of PCR -

Thermocycler which is a laboratory apparatus, used to amplifying the DNA segments in the PCR process. It also animates other temperature sensitive reactions.

Purpose: In this laboratory you will carry out a quantitative real-time PCR (QPCR), using SYBR Green, on the samples that you purified last laboratory session. QPCR serves several purposes in a forensic laboratory and is included in the sample processing workflow for all/most DNA samples. QPCR ultimately measures (either in absolute numbers or relatively) the amount of DNA in a sample and the presence of any inhibitors (either partial or complete). This allows the forensic scientist to determine if more...
