Please list the reagents and apparatus you need in order to perform a restriction reaction in the laboratory.

Homework Answers

Answer #1

Reagents -

1. Restriction enzyme - enzyme which will cut the DNA at a specific site called as restriction site

2. DNA which is to be digested/cut

3. Buffer - which will help to maintain an optimum pH for the working of restriction enzyme

4. Sterile water - to make up a specific final volume

Apparatus -

1. PCR tube - in which all the reagents will be added

2. Microtips and micropipettes - for transferring small volume of the reagents

3. PCR tube holder to keep the PCR tubes

4. Incubator to incubate the reaction mixture at 37℃

Please rate high.

Create a protocol for a restriction digest with a total volume of 30 µl to digest 1.0 µg of plasmid DNA to completion. You have a restriction enzyme in a standard 50% glycerol at 5 Units / µl but wish to do a standard 'overdigestion' in which you will put the maximum allowable enzyme in the digest. (Indicate how many Units will be in the digest.) You have a compatible 10X restriction buffer, and your DNA is at 0.3 µg...

At one point in the lab you will need to perform an acid-base neutralization reaction, using NaOH to neutralize HNO3. Calculate the required amount of NaOH needed for neutralization, knowing that nitric acid is 15.8 M and assuming 6 mL were used. In lab we will use a slight excess (+10%) of NaOH to ensure the acid is neutralized. HNO3 + NaOH à NaNO3 + H2O
