Why do you think there are three primer/probe sets for SARS-CoV-2?

Answer #1

Three primers are

Oligo-dT primers

Random primers and

Sequence specific primers.

As the genome of this viral particle is RNA.

mRNA with poly A tail can be primed with oligo dT primers which have an anchor and bind to the poly A tail of mRNA for genration of complete cDNA from mRNA.

Some Random primers are added which give high cDNA yield. They anneal to all the RNA( tRNA, rRNA, mRNA) and can be used in case of significant secondary structures.

Sequence specific primers for mRNA are Targeting to the specific sequence of viral genome with high sensitivity and specific cDNA pool.

The original TaqMan assay that was deployed by the CDC for SARS-CoV-2 had primer/probe sets to amplify regions specific to SARS-CoV-2 (N1 and N2), a primer/probe set to amplify a region from SARS-CoV-2 and other closely related viruses like the virus that causes SARS (N3), and a primer probe set to amplify a human specific gene (RP). Why was a human gene included?

COVID-19 is caused by infection with the SARS-CoV-2 virus. This virus can infect the type II pneumocytes, a type of cell in the alveoli of the lungs that produce surfactants to ensure proper lung function. Type II pneumocytes have a diameter of about 9 μm. SARS-CoV-2 virus particles have a diameter of about 100 nm. How much larger is a Type II pneumocyte than a SARS-CoV-2 virion? HINT: compare their volumes, assuming both are spheres! The Type II pneumocyte cell...
