. Remembering that a primer/probe set is about 70 bp: What are two regions that would be good targets for a test that would detect ONLY SARS-CoV-2, but not SARS-CoV?

Homework Answers

Answer #1

Although SARS-CoV-2 and SARS-CoV belong to same group of viruses that is corona virus and primer/probe set is only specific for SARS-CoV-2, now commonly known as Covid-10. SARS-CoV interact with one human receptor i.e. ACE, where as SARS-Cov-2 is made up of two region of spike proteins known as S1 and S2, respectively.

S1 region: binding receptor (human or host)

S2 region : Fusion protein to the membrane of cell (human)

So, specific set of primer probe can detect the gene, which encode these proteins and confirm their presence in the patients.

The original TaqMan assay that was deployed by the CDC for SARS-CoV-2 had primer/probe sets to amplify regions specific to SARS-CoV-2 (N1 and N2), a primer/probe set to amplify a region from SARS-CoV-2 and other closely related viruses like the virus that causes SARS (N3), and a primer probe set to amplify a human specific gene (RP). Why was a human gene included?

Using the t distribution table, identify the t statistics that would set the critical regions in a hypothesis test for the following alphas and n’s (2 points each). Remember to check direction and df when choosing the column/row.   One-tailed test, α = .05, n = 10 Two-tailed test, α = .05, n = 10 One-tailed test, α = .01, n = 15 Two-tailed test, α = .01, n = 20
