In SARS-CoV-2 testing using RT PCR involves the use of three primers and probe sets. It is a multiplex reaction where all three primers or probes are used in a single reaction.

Out of three primers, two primers are used to amplify the viral Nucleocapsid gene or N gene and one primers or probe is used to amplify the human RNAse P gene which serves as the internal control which shows that the RNA is isolated from the sample.

Using two primers to amplify the viral gene is used to increase the sensitivity and specificity of the test.
