There is a fundamental difference between the prokaryotic and eukaryotic genome. Whereas the prokaryotic genome is devoid of any intervening sequences or heterochromatin region, the eukaryotic genome contains interspersed heterochromatin or non-coding regions. Thus, the prokaryotic genome contains linear arrangement of open reading frames or genes wheres the eukaryotic genome does not.
Thus, in order to identify ORFs in prokaryotic genome, only the sequence of promoters and start codon are required to be identified. However, very high permutations and simulations are required to identify the ORFs in eukaryotic genome due to presence of introns.
