What would happen to transport into and out of nuclei if Ran-GTP very rapidly degraded into Ran-GDP? What would happen if Ran-GTP could not be converted into Ran-GDP? Please explain your answers.

Answer #1

Ans:- Ran is Ras related nuclear protein (protein of Ras family) involved in transport of protein and RNA during mitosis. inside nucleus Ran GDP converted to Ran GTP by C.C. (regulator of chromosome condensation 1) which is then transported to cytoplasm of cell where Ran GTP get converted in Ran GDP by Ran GAP (Ran GTPase activating factor) and Ran BP1 (Ran binding protein). if Ran GTP degrade very rapidly it will disrupt protein and RNA transport. different concentration of RCC1 and Ran GAP lead hight Ran GTP concentration inside and low Ran GTP Concentration outside of nucleus. if Ran GTP could not be converted into Ran GDP it will increase Ran GTP concentration outside nucleus which disrupt normal ration of Ran GTP and Ran GDP in and out side of nucleus. this disruption in Ran GTP and Ran GDP ratio will interfere in mitosis process of cell cycle.

4. Imagine a G-protein linked receptor that responds to an external signal that tells the cell to undergo cell division. Suppose that there is a mutation so that the G-protein linked to this receptor is unable to degrade the GTP attached to it to GDP. (That is, once the G-protein is bound to GTP, it stays that way, instead of converting the GTP to GDP after time has passed.) What would you predict would happen in the cell? Do you...

What would happen specifically to metabolic pathways and cofactors, etc, involved if the levels/abilities of the glycolytic enzyme, GAPDH, were depleted? like for example I know that Krebs cycle could no longer continue since glucose can no longer be converted to pyruvate and then to acetyl-CoA. What else would be affected? what would increase and decrease?
