Southern blot and PCR confirm the novel amplications in BRCA1 . From a scientific collaborators a genomic DNA from case, suspected carrier of an amplification of exons and two controls were digested using BamH1 and after blotting hybridised using a RT-PCR probe containing exoms. The autoradiogram shows novel fragment of 7kb. The size of the amplified sequences as determined by sequencing is comparable with the size of the extra bands observed after Southern blot analysis, which indicates that the amplified sequences are amplified in tendem.
