Ans 1-Parameters involved in selecting annealing temperature based on Tm of forward and reverse primers are - Lenght and composition of primers . Longer strands have higher melting temperatures, as do sequences with higher G and C content.

Ans 2- Parameters involved in selecting extension time based on the expected Lenght of PCR products are as follows- number of base pairs. Use extension time of 1 minute per thousand base pairs eg. 3 minutes for a 3 kb product. For products less than 1 kb , use 45-60 seconds. Products greater than 3 kb or reactions using more than 30 cycles , may require longer extensions.
