When a ball move upward, it the work done by gravity positive or negative? What about when the ball reaches its the peak, and begin moving downward, what is the work done by gravity?

When a ball move upward, the work done by gravity is always negative

because the ball is displaced (upwards) against (or opposite) the gravitational force (downwards)

But

when the ball reaches its the peak, and begin moving downward then work done by gravity is positive

because in that case the ball moves in the same direction of gravitational force (downwards)
