How do you choose an excitation wavelength for a fluorescence spectrum from an absorbance spectrum?
Fluorescence spectrometer:
In fluorescence spectrometer, first we have to try an excitation
emission scan also called two-dimensional fluroescence, which gives
the total overview for the fluroescence behaviour. Then it allows
to identify separate fluorophors and their relative intensities.
For later identification purposes we can stick to this measurement
because it has the maximal information content. We can quite easily
decide, which excitation/emission wavelength gives you the best
discrimination power.
