How do you choose an excitation wavelength for a fluorescence spectrum from an absorbance spectrum?

How do you choose an excitation wavelength for a fluorescence spectrum from an absorbance spectrum?

Answer #1

Fluorescence spectrometer:

In fluorescence spectrometer, first we have to try an excitation emission scan also called two-dimensional fluroescence, which gives the total overview for the fluroescence behaviour. Then it allows to identify separate fluorophors and their relative intensities. For later identification purposes we can stick to this measurement because it has the maximal information content. We can quite easily decide, which excitation/emission wavelength gives you the best discrimination power.

Problem about double-slit interference. a) Choose and state a wavelength in the electromagnetic spectrum that corresponds to visible light. b)We would like it to be easy to measure the positions of the dark and bright fringes so let’s aim for having a distance of 1cm between adjacent bright fringes. Choose the distance of the screen and the distance between the slits so that this is the case. The slit distance should be much bigger than the wavelength, but much smaller...
