  1. If the PCR reaction does not have the GAPDH band, can you use your data to determine whether that patient has either the swine flu or the coronavirus? Why or why not?

Answer #1

If GAPDH is used as an internal control for the PCR setup, then in the absence of the GAPDH band, we can not use the data obtained to determine if the patient have the disease.

This is because GAPDH is used as internal control and GAPDH is an constitutive gene. Being a constitutive gene, it will be expressed all the time and if proper protocol is followed, GAPDH band will be observed in every human sample tested as they have that gene expressing all the time. This means expression of GAPDH should not be changed by the presence of other infection.

So if the band of GAPDH is not present, it means that we have done some error while handling and preparing the samples. Thus, this data isn't reliable and can't be used.

Question 1 What positive control reaction could you use to be sure that enough good quality DNA was obtained for a successful PCR reaction from your red or pink flower (the untransformed plant)? Note: a positive control gives a positive (not negative) result, so you need a reaction that will give you a PCR band when you use DNA from pink or red carnations. A positive control is important because, impurities in DNA preparations are common and can inhibit Taq...

A full plate of PCR tubes contains 96 wells, each of which can have its own reaction. Determine how much GoTaq Master Mix you ill need to add to your PCR mix, if each reaction requires 12.5 uL and you add on 20% to account for the loss of reagent to plastics. How do you do it?

You have a sample of ssDNA molecule that you will use as a primer for PCR. You dissolved the dried ssDNA peller in TE to a total volume of 400 ul to make a "stock DNA sample." You need to determine the concentration (ug/ul) of this primer DNA in solution. To do this you took 4 ul from the "stock DNA sample" and added these to 996 ul of TE buffer in a cuvette, making a total of 1000 ul...
