Whenever we set the enzymatic reaction we take out enzyme from either -20 degree or 4-degree temperature which is different from the temperature at which the enzyme shows its activity. In case of the substrate, we need to bring it to the temperature at which enzyme can efficiently act on it and catalyse the reaction. Preincubation brings both the enzyme as well as substrate at the optimum temperature of the enzyme where the enzyme starts its activity as soon as we add the substrate
