The use of unconditional replicons which are also known as suicide plasmids that construct the gene duplication and null alleles. Plasmids are conditional to the replication and which can be used to create the defined duplication within the targeted genome. In that type of instances, a specific DNA fragment is placed into the plasmid which is then introduced to a recipient strain which is placed under conditions where the plasmid that cannot replicate. When the plasmid is unable to replicate, the selection for some property of the plasmid like antibiotic resistance maker results in the isolation of plasmid to host chromosome.
