The alpha subunit bind to very upstream sequence of the DNA and then sigma helps in binding with promoter.

RNA polymerase has many subunits like 2 alpha one beta one beta ' one omega which form core enzyme and one sigma which is coenzyme ।when sigma binds the core ENZYME it becomes holoenzyme.

This alpha subunits bind upstream of the promoter and then, sigma subunit binds to the promoter sequences by hydrogen bonding and melt the DNA and start the transcription.

If has to concensus sequences in -10 and. -35 sequences .

In - 10 position, there is TATAAT AND - 35 POSITION , TTGACA.

TRYP AND LAC OPERON IS OPPOSITE IN SENSE THAT LAC OPERON IS INDUCIBLE OPERON WHICH MEANS WHEN LACTOSE IS PRESENT LACTOSE METABOLIZING ENZYME OPERON WILL BE TRANSCRIBED AND WILL METABOLIZE LACTOSE.

BUT IN TRP OPERON WHEN TRYPTOPHAN IS PRESENT , THEN TRYPTOPHAN PRODUCING ENZYME WILL NOT BE PRODUCED.
