we have two drugs used ( Nimesulide and Celecoxib) to inhibit the expression of cox-2 in three cell lines at different concentrations(1,10,20,30,50,70,100) microliter. How to preform the apaptotic essay?

Answer #1

Ans: Protocol used for performing apoptotic assay is explained below-

1. Cells were grown till the logarithmic phase

2. Treat cells with Nimesulide and Celecoxib at different concentrations (1,10,20,30,50,70,100) and incubate for 24 h

3. Harvest cells and centrifuge it

4. Collect cells, wash it using PBS, and resuspend it in 500 µL of 1X binding buffer.

5. Add 5 µL of annexin V-FITC and 5 µL of propidium iodide

6. Incubate cells in dark at room temperature for 5 mins

7. Quantify it using flow cytometry or fluorescence microscopy.

We see that reduction potential and cell potentials are tabulated under standard conditions. We have seen that under non-standard conditions. How do we find the non-standard cell potential when dealing with different temperatures and product/reactant concentrations? What happens to the cell potential when there is far more product than reactant, or far more reactant than product? What happens to the cell potential as temperature is increased?

– During the embryogenesis, axial mesoderm cells are specialized to express different genes. We identified three group of cells present in close proximity. Among these groups one cell subtype (known as Sub1) have probability of 0.5 to express gene XRA. The other cell subtypes (known as Sub2 and Sub3) have a probability of 0.8 and 0.1 respectively. Suppose that we isolated one of these cell subtypes at random and grow them in the lab to study gene expression patterns. We...

2. Which of the following statements about epidermal growth factor (EGF) is false? (a) EGF stimulates cell division after an injury (b) EGF is a local signal, meaning that it binds to target cells that are close to the site where it was secreted (c) The EGF receptor is the same receptor that is used by insulin, but it is expressed in a different location (d) A mutation that causes the EGF receptor to become overactive can cause cancer (e)...

Comparing two drugs . Makers of generic drugs must show that they do not differ significantly from the “reference” drugs that they imitate. One aspect in which drugs might differ is their extent of absorption in the blood. We have some data taken from 20 healthy nonsmoking male subjects for one pair of drugs. This is a matched pairs design. Numbers 1 to 20 were assigned at random to the subjects. Subjects 1 to 10 received the generic drug first,...

Define and explain how two of the following three methods were used (or not used) by the consumers to estimate the overcharge in the lysene cartel: before and after, cost-based and yardstick. [We did not discuss the third in class, though it is about a different type of comparison than the before and after method.]
