Affinity is a measure of the tightness with which a drug binds to the receptor. It is a passive process. Affinity is how avidly a drug binds to an open receptor or how the chemical forces that cause a substance to bind its receptor. It is like a drug's desire to connect an open receptor. It refers to how much attraction there is between a drug and a receptor, like a magnet and metal. Some drugs have higher affinity and others have a lower affinity for the same binding sites.

Efficacy is used to refer both to the maximum response achievable from a pharmaceutical drug in research settings, and to the capasity for sufficient therapeutic effect or beneficial change in clinical settings. Efficacy refers to whether a dug demonstrates a health benefit over a placebo or other intervention when tested in an ideal situation, such as tightly controlled critical trail. These studies focus on primary parameter to be shown statistically different between placebo and intervention groups.
