2a. Predict each of the folloiwng experimental changes will do to the initial rate of remonal of the phosphate group from Protein X catalyzed by alkaline phosphatase. Answer each with increase decrease or no change and explain biochemically why.
1) increase the temperature of the reaction mixture from 25 degrees C to 37 degrees C
2) Change the concentration of Protein X from a value equal to 1Km ti a value equal to 2 Km
3)Increase the reaction temperature from 37 degrees C to 100 degrees C
4) decrease the pH of the reaction mixture from pH=9 to pH=1
1) at 37 degree the phosphate activity will be at peaks due to proper structural confirmation and stabilty of protein
2) activity of the phosphatase will increse upto the saturation level of it and after that there will be no change in the reaction rate and the activity will be constant due to available sites of enzymes are completly filled no other protein will be involved no reaction
3) the protein will be denatured at 100 degrees as a result the activity will decrease
4) biochemical activty will decrease due to unstabilty in the protein folding and charge difference occured due to change in the pH, the optimal pH for activity of alkaline phosphatase is 9
