A TLC is run using 2:1 hexane/ethyl acetate on an alumina plate. There were three spots all close to the bottom of the plate with low Rf values. How could the TLC be changed to increase the Rf's of the three spots to get better separation?

Answer #1

Different compound in the sample mixture travel at different rate due to difference in their attraction to the stationary phase. If you take silica as a statiomsta phase then since silica is polar it will bind steongly with polar compound while non polar compound bind less and trabet more higher uper in plate.

Regarding your problem please increase the polarity of mobile phase means you have to increase the amount of ethylacetate so that polarity increase and 3 distinct spot seperate easily.

If a compound has an Rf of 0.25 when a 1:1 mixture of hexane and ethyl acetate is used as the solvent on a silica gel TLC plate, will the Rf value be larger or smaller if a 4:1 mixture of hexane and ethyl acetate were used as the solvent? Ethyl acetate is another common organic solvent used in the type of TLC experiment you performed: it has a higher molecular weight and boiling point than either petroleum ether or diethyl ether, and is more polar than them as well. That is, predict how the behavior...

I know that 1:4 acetate; hexane eluting solvent is nonpolar. So what about the 1:2 acetate; hexane eluting solvent? Is it polar or non polar? When I used 1:4 acetate; hexane eluting solvent, I got that the starting material was more polar from the product because it has a smaller Rf value? If the eluting solvent changed from 1:4 acetate; hexane to 1:2 acetate; hexane, would the Rf values get larger? smaller? same? Explain why please

When ACE, ASP and CAF were analyzed by TLC using 2:9:9 mixture of t-butyl alcohol, 4-methyl-2-pentanone and ethyl acetate respectively the following Rf values were obtained: ACE (0.51), ASP (0.15) and CAF (0.35). These results are not the same ones obtained when using 2% acetic acid and 98% ethyl acetate as the mobile phase? The TLC chromatogram shows the result for a separation of a mixture of three compounds using a solvent of medium polarity. Only two spots are visible because two compounds did not separate. If spot A represents a mixture of two compounds, would you be able to separate the two components by decreasing or increasing the solvent polarity? Please explain. Sketch the TLC’s that you would expect to see if you were to develop a TLC in a) a polar solvent,...
