c) A final step in the analysis of enzymatic activity during Enzyme Practical I: Effect of time, pH and temperature on the activity of acid phosphatase and Enzyme Practical II: Kinetics, effect of varying substrate concentration on enzyme activity was the addition of NaOH to your samples. Provide two reasons why you added NaOH and explain the molecular effects that NaOH has in each case!

Answer= NaOH is added it has effects

1] NaOH converts p-nitrophenol to p-nitrophenolate and developes yellow color.

2]Acid phosphatase hydrolyses phosphate ester bonds under an optimum pH of 4.8 & optimum temperature of 370C.

in this experiment we add artificial substrate para-nitrophenylphosphate which will be hydrolysed by acid phosphatase,releasing p-nitrophenol and inorganic phosphate(Pi)

Both   para-nitrophenylphosphate and p-nitrophenol are colorless at acidic pH values addition of NaOH will stop the reaction due to increase in pH of the reaction medium.
