The plant toxin retinone blocks oxygen consumption by mitochondria. Addition of succinate restores oxygen consumption. Which of the proton pumps of oxidative phosphorylation must be inhibited by retinone, therefore? Explain your answer.

Answer #1

ANSWER: Rotenone is a broad spectrum insecticide and pesticide.

Mechansim of action of Rotenone: It interferes with the electron transport chain in the mitochondria. It inhibits Fe-S complex in the Complex-1 to the mobile electron carrier that is Ubiquinone.

Complex-1 is unable to pass the electrons to CoQ which results in accumulation of Oxygen in the mitochondrial matrix. This cellular oxygen is reduced to the free radicals which consequently result in the damage to DNA and other components of mitochondria.

Mitochondria produce most of the cell's ATP by oxidative phosphorylation. Electrons from NADH and FADH2 formed in the TCA cycle are transported through the electron transport chain (ETC) which consists of four complexes (I, II, III and IV). Hereby, a proton gradient is formed across the inner membrane of the mitochondria which is used by the ATP synthase to form ATP. In isolated mitochondria, oxygen consumption rate (OCR) and rate of ATP synthesis can be determined, respectively. Seahorse and NOVOstar...

Background Almost all animal and plant cells require oxygen and give off carbon dioxide. This is called gas exchange and it occurs by passive diffusion. Some animals do all or most of their gas exchange across their skin (for example amphibians) but they have very low metabolisms and use very little oxygen and get rid of very little carbon dioxide. On the other hand humans (and other mammals) have very high metabolic requirements and we can’t rely on gas exchange...
