You forgot about your disk diffusion test plate that was supposed to incubate at 35˚C for 48hrs and now it’s been at 35˚C for several weeks. What do you think would happen if you let the KirbyBauer test plate continue to incubate for that long?
As kirbybauer plate is used for antibiotic sensitivity when incubated for 2 days the bacteria which are resistent to antibiotic survive and others dies but if we incubate it for weeks then if the bacteria is not resistant then no growth will occur but I may produce new strain which are resistant to that antibiotic whose growth will be much more but after some time the food in the culture will be lost so the bacterial growth will fall at a hugh rate and the result for antibiotic suseptibiliy would not be found as bacteria may die due to antibiotic or due to end of food
