The human H1 histone gene lacks introns and its immune to nonsense mediate decay. Design an experiment to test the hypothesis that the H1 gene immunity to non-sense mediate decay is because it lacks a introns. Be sure to include a control in your experiment. What results would support the hypothesis that H1 histone mRNA's immunity to NMD is due to its lack of introns. What results would support the hypothesis that its immunity is due to other elements of the H1 histone gene.

Answer #1

Answer- The hypothesis can be tested by using a vector in which the DNA sequence without having introns should be inserted and it should also have premature stop codon in the sequence. Another vector for control should be made by insertion of DNA sequence having introns and premature stop coding.

The supportive results will show the expression of the sequence without introns but no expression in case of control.

If the control also show expression of the sequence then it can be concluded that the controlling agent for immunity to NMD is some other element.
