  1. Several compounds known as uncouplers function to uncouple electron transport from ATP synthesis in the mitochondria. These uncoupling compounds include 2,4-dinitrophenol, dicumarol, and FCCP. These agents transport hydrogen ions from the intermembrane space of the mitochondria into the mitochondrial matrix. Explain how this function uncouples electron transport and ATP synthesis.

Homework Answers

Answer #1

Mitochondria is the store house of energy where ATP is synthesized by a process known as electron transport chain and this is a process by oxidative phosphorylation by using 3 protons in the intermembrane to generate one ATP and this process is inhibited by uncoupling compounds like 2,4-dinitrophenol,dicumarol,and FCCP .This uncouplers that disrupts the oxidative phosphorylation in mitochondria by dissociating reactions of ATP synthesis in the electron transport chain. They inhibit the coupling between electron transport and phosphorylation and inhibits ATP syntesis.

Mitochondria produce most of the cell's ATP by oxidative phosphorylation. Electrons from NADH and FADH2 formed in the TCA cycle are transported through the electron transport chain (ETC) which consists of four complexes (I, II, III and IV). Hereby, a proton gradient is formed across the inner membrane of the mitochondria which is used by the ATP synthase to form ATP. In isolated mitochondria, oxygen consumption rate (OCR) and rate of ATP synthesis can be determined, respectively. Seahorse and NOVOstar...
