What is Km? Write Km in terms of the rate constants from the H-M-M model of enzyme- catalyzed reactions? What is Km a measure of? How can we use Km to determine the fraction of enzymes that are actively processing substrate into product? (Note: you will not be given this equation on an exam. This is something you must know). Km is the concentration of reactant or substrate at which half of the active site of enzyme are filled

It measure the affinity of enzyme towards substrate

Km with high value indicates that the enzyme has low affinity thus require more substrate and with low value indicates that enzyme have high affinity towards substrate convert them into product easily

in a plot

intercept = 1 / Vmax

gradient = Km / Vmax

x intercept = -1/ Km

So we can determine km easily by dividing gradient by intercept
