Wild type is used as control because the other strains are mutant. at a different point in the biosynthetic pathway.
So they not grown in the minumal media but wild type will grow. So it used as positive control.

Streak used to isolated single cells from growing colonies. technique is to dilute the cells on the surface of the agar in successive, overlapping streaks, until individual cells are separated or distributed to form isolated single colonies.

Minimal medium contain minimum nutrients without the presence of specific limiting growth factor. So mutant strain not able to grow here only wild type will grow in the minimal media.
