LDH is found in many of the body's tissues and organs , including the muscles, liver, heart, pancreas, kidneys, brain and blood cells. The LDH test is mainly used to help identify the location and severity of tissue damage in the body.
LDH is a stable enzyme, present in all cell types, which is rapidly released into the cell culture medium upon damage of the plasma membrane. LDH is the most widely used marker to run a cytotoxicity.
The method of LDH is based on flurometric determination of enzyme kinetics when L - lactate is converted to pyruvate.
A substrate concentration of 35mM in the reaction mixture was considered to be optimal for the LDH assay.
