Explain how the extraction with chelating matrix and protease (InstaGene Matrix) occurs?

Homework Answers

Answer #1

Extraction of DNA with InstaGene Matrix can be performed in following steps:-

1. Cell sample with the help of sterile pipet tip are transferred into a microtest tube containing InstaGene Matrix. This matrix is made up of negatively charged microscopic beads called "chelates". It is named chelate because of ability to grab metal ions out of solution reqired as cofactors in enzymatic reactions.

2. Sample is heated to lyse the cell so that their cellular constituents will be separated including lysosomes which contain powerful DNAase which can digest our DNA of interest released by cell sample. Here the main purpose of InstaGene Matrix in DNA extraction is to adsorb cofactors like Mg2+  make it unavailable for enzymes. This bolcks all enzymes degradation of the extracted DNA and result in population of intact genomic DNA which can be used as template for PCR reactions.
