So when purifying this PFK-1 from the rat liver the total protein content will be unchanged or stay constant,as the PFK-1 does not have any effect in the proteins.
The activity of the PFK-1 depends on the ATP/AMP ratio,the activity of the PFK-1 is inversely related with the ATP/AMP ratio.So at the time of purification the ratio of ATP/AMP is generally low in concentration,so the activity of the PFK-1 may increase,now at this point if the activity gets increased there is a possibity lies there that the specific activity will also increase because the specific activity is the activity of an enzyme per miligram of total protein.
