1 In UAS- GAL4 system , GAL4 is the transcriptional activator which binds to the enhancer sequence UAS , then there is Transcription of the downstream coded genes like RNAi. If there is no GAL4 , there will be no Transcription. That's why RNAi not working here.

2. Chi square test is used to compare the observed values to the expected ones . And to know whether the difference observed is significant or not .

The p value indicates whether the difference is significant or not. When p value is 0.0 5 or less than it , the difference is significant.
