The liver plays a critical role in maintaining glucose homeostasis. It is therefore not surprising that insulin regulates hepatic glucose production (HGP) through multiple mechanisms. HGP is inhibited by intracellular signals resulting from the direct binding of insulin to its hepatic receptor, resulting in a rapid reduction in glycogenolysis. In addition, insulin can indirectly suppress HGP by several means, including the reduction of circulating free fatty acids (FFAs) and glycerol as a result of the inhibition of lipolysis, the suppression of glucagon secretion and by means of a change in neural input to the liver secondary to insulin action in the brain.
