separation by centrifugation is influenced by two factors-

1-RCF

2-centrifugation time.

in centrifugation urine should be centrifuged within normal time limit on given RCF. if it is centrifuged for longer time on high RCF then it is observed that higher recovery of microrganism is seen.recovery rate under sediments ranged for below 50 to 80 percent. increased recovery is not proportional to RCFand time but certainly increases.

centrifugation for prolonged time also leads to cell rupture and cast fragmentation.

capping the tubes during the centrifugation is very important so that fluid or any material does not spill or come out of thr tube while rotating on high RPM.

it is important for the safety of centrifuge machine and also for the reasercher and environment if tube contain any Hazardous material.
