How is the regulation of phosphofructokinase-2/fructose 2,6-bisphosphatase-2 affected by Type 1 diabetes?
Fructose bisphosphatase 2 is an enzyme od EMP pathway which directly regulates the rate of glycolysis pathways. It regulates a reversible reaction in EMP patheaway by conversion and degradation of fructose 2,6 bisphospjate from fructose 6 phosphate. And this stage is rate determining step of the pathway.
Insulin activates a protein phosphatase which dephosphorylates the phosphofructokinase-2 complex and causes favored enzymatic activity. Phosphofructokinase 2 then increases production of phosphofructokinase 2. it activates glycolysis and inhibits gluconeogenesis. In presence of diabetes mellitus 1 there is insulin defecient condition so this regulation is hampered and there is no PFK2 increased production, this will inhibit glycolysos and amount of blood sugar starts to increase
