Once the smear is stained with carbol fuschin to differentiate acid fast bacteria from non acid fast bacteria, the acid fast bacteria will appear in red colour and the background( non acid fast bacteria) will be decolourised. Then counter stain the smear with methylin blue or brilliant green.The acid fast bacteria will remain as red and the background ( the decolourised bacteria) will uptake the colour, such as in blue colour if methyline blue is used,or in green colour if brilliant green is used.Thus the bacterias can be easily distingushed
