what is the principle of the glucose oxidase method ?

what is the principle of the glucose oxidase method ?

Homework Answers

Answer #1

GLUCOSE OXIDASE METHOD

  • based on colorimetry
  • glucose oxidase catalyses the oxidation of glucose to gluconic acid
  • hydrogen peroxide is detected by chromogenic oxygen acceptor phenolaminophenazone
  • intensity of color depend on glucose conc

trinder method-

alpha - D- glucose ---------mutarotase -------> beta-D-glucose

beta-D- glucose + H2O + O2 -----------GLUCOSE oxidase ------> D-gluconic acid + H2O2

H2O2 + 4 aminophenazone + phenol ------peroxidase-----> quinoemine + 4 water

requirement for test

  • blood sample
  • photometer
  • cuvette
  • water bath
  • tets tube
  • pipette

blood normal range

  • fasting -70-110mg/dl
  • postprandial - 110-140mg/dl
  • renal thrashold 180mg/dl
