  • based on colorimetry
  • glucose oxidase catalyses the oxidation of glucose to gluconic acid
  • hydrogen peroxide is detected by chromogenic oxygen acceptor phenolaminophenazone
  • intensity of color depend on glucose conc

alpha - D- glucose ---------mutarotase -------> beta-D-glucose

beta-D- glucose + H2O + O2 -----------GLUCOSE oxidase ------> D-gluconic acid + H2O2

H2O2 + 4 aminophenazone + phenol ------peroxidase-----> quinoemine + 4 water
